For the biscuit mix: In large bowl, combine flour, sugar, and salt. Whisk until well combined. Set fine-mesh strainer over bowl. Add buttermilk powder, baking powder, and baking soda to fine-mesh strainer and tap side of strainer to sift mixture into bowl.
Whisk until very well combined, about 1 minute. Transfer biscuit mix to large airtight storage container. (Biscuit mix can be stored at room temperature for at least 2 months.)
To make one batch of biscuits: Adjust oven rack to middle position and heat oven to 450 degrees. Line rimmed baking sheet with parchment paper.
In medium bowl, whisk water and melted butter until combined.
Add 2⅓ cups Buttermilk Biscuit Mix and use rubber spatula to stir until just combined and no dry mix is visible.
Spray inside of ¼-cup dry measuring cup with vegetable oil spray. Use greased measuring cup to scoop batter and use rubber spatula to scrape off extra batter. Drop scoops onto baking sheet to make 10 to 12 biscuits (leave space between biscuits and respray measuring cup as needed).
